炭疽桿菌是炭疽病的病原體。炭疽桿菌產(chǎn)生的主要毒力因子是γ連接的聚-D-谷氨酸膠囊和由三種成分組成的外毒素，保護(hù)性抗原（PA），致死因子（LF）和水腫因子（EF）。這些蛋白質(zhì)不是單*毒的，而是以二元組合起作用。PA（細(xì)胞結(jié)合成分）與LF（酶部分之一）的復(fù)合物被稱為“致死毒素”并可導(dǎo)致死亡。PA和酶促EF一起引起皮膚水腫。分泌后，PA被膜肽酶切割，使63kDa羧基末端片段寡聚化成七聚體。切割是暴露EF和LF的結(jié)合位點(diǎn)的必要步驟。組裝的PA復(fù)合物和相關(guān)的LF或EF通過(guò)胞吞作用進(jìn)入細(xì)胞。PA介導(dǎo)LF和EF向細(xì)胞質(zhì)的轉(zhuǎn)移，其中這些酶識(shí)別并蛋白水解其靶標(biāo)。LF組分是一種鋅依賴性金屬蛋白酶，可裂解絲裂原活化蛋白激酶激酶（MAPKK），從而削弱其功能，從而阻斷MAPK信號(hào)通路。EF是鈣調(diào)蛋白依賴性腺苷酸環(huán)化酶，其在細(xì)胞中產(chǎn)生過(guò)量的環(huán)AMP，導(dǎo)致水腫。目前的研究集中于開發(fā)有效的疫苗以及篩選LF酶活性的有效抑制劑。

炭疽保護(hù)性抗原片段（PA 20）
從炭疽芽孢桿菌
保護(hù)性抗原（PA）是炭疽毒素的受體結(jié)合成分。原始的83kDa蛋白質(zhì)被細(xì)胞弗林蛋白酶樣蛋白酶切割，形成活化的63kDa單體PA63。在這種形式中，PA63與其他PA63分子結(jié)合形成環(huán)狀七聚體。PA63還具有對(duì)于與細(xì)胞表面上形成的孔相鄰的酶LF和EF特異性的結(jié)合位點(diǎn)，用于易位。對(duì)于醉酒細(xì)胞，PA應(yīng)與EF或LF一起使用約7倍過(guò)量。
